Use Skills Wisely
Though you can level them up and augment them as the game goes on, your hero has three main skill varieties: a whirling slash attack, a leaping slam attack, and the ability to summon a clone of himself to do extra damage.
While you don't spend any mana or gold or anything to use them, each skill does have a cooldown, so strategic use of the skills is necessary when the game gets tough.
The Whirl skill has the quickest cooldown of all of your skills, so feel free to channel your inner Link and spin attack to your heart's content. The Leap skill has a significantly longer cooldown, however, and should be saved for times where you're either in a pinch, or there is a large group of enemies clustered together out of your attack range.

It's especially useful in closing the distance against deadlier long-range enemies when a melee enemy is blocking your path. The Clone skill has the longest cooldown of all, but oddly, the cooldown starts when you cast the spell, not when your clone disappears. Since your clone actually hangs around for a while, this means that the effective cooldown of the move, between your clone disappearing and the time at which you can use the skill again, is around 3 seconds. This generally means it's advisable to have a clone out pretty much all the time.
A Note On Runes
As you progress through the game and level up, you will unlock runes that can augment your skills. The most important thing to remember in regards to these runes is that you can swap them out at any time, so the best way to use them is often situational. Don't be afraid to look at a boss's skill set and swap out runes based on what you think will be most effective. Between bosses, however, I personally prefer to have runes that boost gold collection equipped.
G.R.E.A.M. (Gold Rules Everything Around Me)
The easiest way to strengthen your character in Nonstop Knight is to purchase equipment upgrades with the gold that enemies drop, so make sure that you keep all your gear appropriately leveled. Occasionally, enemies will drop stronger gear, so don't hesitate to sell your old gear after equipping better items.
In addition, your hero is gathering gold for you even if you're not playing, so if you feel like you're stuck or if you really want to level that sword up, put the game down for an hour or two and by the time you come back, there'll be gobs of gold waiting for you.
Ascend Carefully
Ten floors into the game, your hero will tell you that you should Ascend, since it awards you tokens that can be used to purchase permanent upgrades to your character. He'll also tell you that this will wipe out all the loot that you've collected so far.
What he won't tell you is that if you don't Ascend right away, your reward for Ascending will increase little by little. So if you are able to Ascend, and your hero is plowing through enemies and bosses with ease, wait until you're having a bit more trouble and then cash in. It'll be worth the wait.
When Things Get Hairy..
So you're having a bit of trouble on a floor and need some help. There are 2 things you can do. Either quit and leave your hero to his own devices, or simply open up the upgrade screen and give yourself some time to think and devise a strategy. Although the game never pauses, opening upgrade screens puts the game into slow-motion, and although it doesn't really give your character any advantage, it can actually be helpful and allow you to regroup, upgrade, and plan out your skill usage (or simply Ascend, if you can).
At the end of the day, Nonstop Knight is definitely more of a clicker game than it is an action-RPG -- so as I said before, if you're having trouble, you can literally put the game down for a while and come back to it later. When you do, your hero will have more gold, and you'll be able to advance further.
